Bu komplikasyonlar benzer biçimde deneysel diyabet modellerinde de oluşmaktadır. Bu çalışmada, streptozotosin ile oluşturulan altı haftalık diyabetin sıçan uterus düz kasında asetilkolin ve prostaglandin F2α ile oluşturulan kasılmalar üzerindeki etkilerinin incelenmesi amaçlanmıştır. Streptozotosin injeksiyonundan 3 gün sonra kan şekerleri ölçülen dişi sıçanlar altı hafta süre ile metabolik kafeslerde tutulup tükettikleri yem ile su miktarları ölçülmüş ve vücut ağırlıklarındaki değişimler izlenmiştir. Bu arada hayvanların çıkardıkları feçes ve idrar miktarları da takip edilmiştir. Streptozotosin injekte edilen hayvanlarda kan şekerinin yüksekliğinin yanısıra, polifaji, poliüri ve polidipsi gibi tipik diyabet semptomları gözlenmiştir. Altı haftanın sonunda diyabetik hayvanlardan alınan kan numunelerinde glukoz, kolesterol ve HDL-kolesterol düzeyleri ölçülmüş ve kontrole göre istatistiksel anlamlı farklılıklar saptanmıştır. Kontrol ve diyabetik hayvanlardan uterus dokuları çıkarılarak tartılmış ve izole organ banyosunda asetilkolin ve prostaglandin F2α cevapları incelenmiştir. Streptozotosin diyabetik sıçanların uteruslarındaki kontraktil asetilkolin ve prostaglandin F2α cevaplarının kontrollere göre istatistiksel anlamlı biçimde düşük olduğu bulunmuştur. Similarly, these complications may occur in the experimental models of diabetes. In the present study, it was aimed to investigate the effects of six weeks of diabetes, induced by streptozotocin, on the contractions of rat uterus elicited by acetylcholine and prostaglandin F2α. Female rats whose blood glucose levels were measured three days after the injection with streptozotocin were housed in metabolic cages for six weeks, and, during this period, their food and water consumptions and changes in their body weights were monitored. Urine and faeces amounts of animals were also recorded. In addition to high blood glucose levels, typical symptoms of diabetes such as polyphagia, polydipsia and polyuria were observed in animals injected with streptozotocin. At the end of six weeks period, glucose, cholesterol and HDL-cholesterol levels were measured in blood samples from diabetic animals and statistically significant changes were detected in these parameters comparing to controls. Uterus tissues from diabetic animals and their controls were weighed after their excision and their responses to acetylcholine and prostaglandin F2α were examined in the isolated organ bath. The contractile responses of streptozotocindiabetic rat uterus to acetylcholine and prostaglandin F2α were found to be decreased significantly relative to those obtained from controls. |
